The mucus layer

A protective coat of mucus sits on top of the cells, keeping most microbes at arm’s length from the actual barrier. A healthy mucus layer is your first buffer.

Tight junctions

The epithelial cells are stitched together by protein complexes called tight junctions, built from proteins with names like ZO-1, occludin, and claudin. Think of them as the grout between tiles. When the grout is intact, nothing slips through the cracks. When it loosens, the wall becomes leaky.

The microbiome changes. The community of trillions of bacteria living in your gut, your microbiome, tends to lose diversity with age. The immune cells turn up the heat. Research shows that aging itself reprograms the immune cells in the gut wall toward a more inflammatory state. Studies of the aging colon find elevated populations of pro-inflammatory innate immune cells, including neutrophils, dendritic cells, monocytes, and macrophages, along with higher inflammatory cytokine output. One study found that monocyte-driven inflammaging actively reduces intestinal barrier function, an effect that appeared especially pronounced in females. This creates a vicious cycle: a weaker barrier feeds inflammation, and that inflammation further weakens the barrier.

The result is what researchers call increased microbial translocation, a polite way of saying that bacterial fragments such as lipopolysaccharide (LPS) slip across the leaky wall into circulation. The immune system treats these fragments as a threat and mounts a low-grade response, day after day. That is inflammaging, lit and fed from the gut.
